Sin embargo, los factores de estr\u00e9s solo se pueden detectar despu\u00e9s de que los s\u00edntomas son visibles. Ser\u00eda beneficioso detectar los problemas potenciales de forma m\u00e1s temprana: cuanto antes se pueda abordar un problema de producci\u00f3n, menor ser\u00e1 su impacto.<br><\/span><\/p>\n\n\n\n<figure class=\"wp-block-image aligncenter\"><img decoding=\"async\" src=\"https:\/\/endowment.org\/wp-content\/uploads\/imported-files\/Research-Figure1.jpg\" alt=\"\" class=\"wp-image-15825\"\/><\/figure>\n\n\n\n<figure class=\"wp-block-image aligncenter\"><img decoding=\"async\" src=\"https:\/\/endowment.org\/wp-content\/uploads\/imported-files\/Research-Figure2.png\" alt=\"\" class=\"wp-image-15826\"\/><\/figure>\n\n\n\n<p class=\"wp-block-paragraph\"><span style=\"color: #000000;\">Nuestra investigaci\u00f3n utiliza un sistema de im\u00e1genes multiespectrales para determinar la intensidad de la fluorescencia de la clorofila (CFI). Todos los organismos fotosint\u00e9ticos contienen clorofila, que emite fluorescencia. Esto significa que una peque\u00f1a fracci\u00f3n de la luz que absorben las plantas se reemite en forma de luz roja y roja lejana. S\u00ed, las plantas brillan, literalmente. No podemos verlo porque la fluorescencia es demasiado d\u00e9bil (solo entre el 1 y el 2% de la luz que incide sobre la planta), pero es f\u00e1cil de fotografiar. Iluminamos las plantas con LED azules y, a continuaci\u00f3n, tomamos una fotograf\u00eda a trav\u00e9s de un filtro que solo permite que la luz roja y la luz roja lejana pasen a trav\u00e9s del objetivo hasta el sensor de imagen. El resultado: una imagen de la luz producida por la planta. Tambi\u00e9n tomamos fotograf\u00edas de las plantas bajo luz roja e infrarroja y utilizamos estas im\u00e1genes para calcular el \u00cdndice de Vegetaci\u00f3n de Diferencia Normalizada (NDVI, un \u00edndice habitual del vigor o el verdor de las plantas). Hemos observado que un mayor aporte de fertilizante da lugar a valores m\u00e1s altos de CFI y NDVI, as\u00ed como a valores de NDVI m\u00e1s uniformes en todo el dosel (figura 1).<\/span><br><span style=\"color: #000000;\">&nbsp;<\/span><br><span style=\"color: #000000;\">Nuestro sistema de im\u00e1genes tambi\u00e9n muestra el potencial de detectar estr\u00e9s por temperatura. Nuestra investigaci\u00f3n beneficiar\u00e1 a la industria mediante el desarrollo de m\u00e9todos de edici\u00f3n gen\u00e9tica para la mejora de cultivos florales. Se est\u00e1 examinando la capacidad de conferir resistencia a enfermedades mediante la tecnolog\u00eda CRISPR en cultivares de gerbera del mercado, que no son un genotipo modelo. En el \u00faltimo a\u00f1o, nuestros esfuerzos se concentraron en cuatro \u00e1reas: multiplicaci\u00f3n de brotes, inducci\u00f3n de organog\u00e9nesis y embriog\u00e9nesis som\u00e1tica, transformaci\u00f3n gen\u00e9tica y dise\u00f1o de constructos CRISPR.<\/span><\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><span style=\"color: #000000;\">Disparar Multiplicaci\u00f3n<\/span><\/strong><br><span style=\"color: #000000;\">Hemos determinado con \u00e9xito el medio de cultivo de tejidos \u00f3ptimo para la multiplicaci\u00f3n de brotes de los cultivares de gerbera \u2018Flori Line Maxi Yellow\u2019, Revolution \u2018Bicolor Red Lemon\u2019, Garvinea \u2018Sweet Love\u2019 y Majorette \u2018Pink Halo\u2019, lo que nos proporcion\u00f3 una fuente de l\u00e1minas foliares y pec\u00edolos para experimentos de cultivo de tejidos y transformaci\u00f3n.<\/span><\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><span style=\"color: #000000;\">Organog\u00e9nesis y Embriog\u00e9nesis Som\u00e1tica<\/span><\/strong><br><span style=\"color: #000000;\">Para producir plantas transg\u00e9nicas y editadas gen\u00e9ticamente, se requieren m\u00e9todos eficientes para regenerar brotes de gerbera a partir de cultivo de tejidos. Tambi\u00e9n estamos investigando la embriog\u00e9nesis som\u00e1tica, un enfoque alternativo para la regeneraci\u00f3n, debido a que el tejido embriog\u00e9nico es un buen objetivo para la biol\u00edstica (la entrega directa de fragmentos de ADN en las c\u00e9lulas mediante una pistola g\u00e9nica) y la edici\u00f3n gen\u00e9tica sin transgenes. Con estos fines, pudimos determinar el tratamiento de inducci\u00f3n de brotes m\u00e1s eficaz para pec\u00edolos y limbos foliares de Gerbera \u2018Flori Line Maxi Yellow\u2019 y Majorette \u2018Pink Halo\u2019, y se indujo callo potencialmente embriog\u00e9nico a partir de Majorette \u2018Pink Halo\u2019 con 2,4-D.<\/span><\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><span style=\"color: #000000;\">Transformaci\u00f3n gen\u00e9tica y constructos CRISPR<\/span><\/strong><br><span style=\"color: #000000;\">Para determinar c\u00f3mo introducir genes deseables (en este caso, resistencia al o\u00eddio), utilizamos lo que se conoce como genes marcadores. Se trata de genes for\u00e1neos introducidos cuya expresi\u00f3n proporciona un rasgo f\u00e1cilmente identificable que nos permite saber que una c\u00e9lula determinada puede ser transformada. Hay dos tipos de genes marcadores: los marcadores de selecci\u00f3n (com\u00fanmente de resistencia a antibi\u00f3ticos o herbicidas) y los marcadores visuales (fluorescencia o coloraci\u00f3n azul). Los marcadores de selecci\u00f3n matan a todas las c\u00e9lulas excepto a las transformadas, mientras que los marcadores visibles ofrecen una confirmaci\u00f3n visual de la transferencia g\u00e9nica. Descubrimos que el gen de la higromicina B fosfotransferasa (HPH) era el mejor marcador de selecci\u00f3n para los cultivares con los que trabajamos, en comparaci\u00f3n con otros marcadores de selecci\u00f3n utilizados previamente en la literatura. Adem\u00e1s del marcador HPH, introdujimos un transg\u00e9n de prote\u00edna verde fluorescente (GFP, un marcador visual com\u00fanmente utilizado para indicar la expresi\u00f3n de transgenes) en explantes foliares de \u2018Flori Line Maxi Yellow\u2019 utilizando *Agrobacterium tumefaciens* C58 en una serie de experimentos multifactoriales. La expresi\u00f3n de GFP pudo observarse en m\u00faltiples sitios a lo largo de nuestros explantes foliares despu\u00e9s de dos semanas (como lo indica la brillante fluorescencia verde en la figura 2), lo que indica una transformaci\u00f3n estable. En presencia de 0,1 mg\/L de IBA y 2,0 mg\/L de BAP, se obtuvo tejido en regeneraci\u00f3n que expresaba GFP.
